To clean the mating surface between the differential flange and the companion flange, you CAN do it without removing the wheel. I would suggest you set the rear up on jackstands securely, use the e-brake to brace the wheels while you loosen the bolts. Then, it may be easier to move the axle around if you jack the wheel back up to load the suspension. This will give the loosened end of the shaft more wiggle room.
To get to the other end, yes, you must remove the wheel. As you can see, the splined shaft is threaded on the end, and the wheel hub nut bolts to the end of it. Unless you take the wheel off, the nut, and the hub, you will not be able to get to the splines at the end of the axle.
