Power loss

Im only running on 5 cylinders how much power difference does that 1 cylinder make?

I would say more than 1/6th - it disrupts the motion and creates a syncopation in the rotation that can sap power (destructive waveform). You would have to dyno it to get a good idea, but that is moot - don't drive it with only one cylinder.

Driving it with no plug and a working fuel injector is not a smart idea. Much of the fuel will be ejected from the spark plug hole. If you leave the plug in you'll wash down the cylinders with unburnt fuel. If you continue to drive the car you will end up causing some damage.
